Su Nature Biotechnology, Ron Cheng e altri partecipanti al Resilience Project lanciato dalla fondazione Sage BioNetworks due anni fa, analizzano (e spesso ricontrollano in vitro) 12 data-set privati e pubblici di genomi, 400 mila dei quali appartengono a 23andMe:
A comprehensive screen of 874 genes in 589,306 genomes led to the identification of 13 adults harboring mutations for 8 severe Mendelian conditions, with no reported clinical manifestation of the indicated disease.
In quella montagna di sequenze, hanno cercato 874 geni che quando sono mutati causano 584 malattie genetiche – neurologiche, metaboliche ecc. – che si dichiarano nell’infanzia, e che non erano state diagnosticate nei donatori, arrivando così a soltanto 16 mila genomi. Il primo problema è che
more than 75% of the candidates were eliminated due to inaccurate or low-confidence variant calls in the existing data,
dice Stephen Friend, co-ideatore del Resilience Project, chissà come sarà contenta la CEO di 23andMe! Il secondo problema è ancora più frustrante:
none of the 13 final candidates could be contacted with follow-up questions due to limitations in the original studies’ informed consent policies. It will be impossible to determine whether these people are truly resistant to disease without additional information.
Che cosa li ha protetti? Risposta in un’altra puntata. Nel frattempo su Nature e altrove ci sono commenti pieni di promesse circa una via alternativa per la ricerca di terapie. Hmm… da un lato i 13 “super-eroi” genetici sono stati “resilienti” fino al sequenziamento del loro genoma, dopo non si sa. Dall’altro sono 25 anni che si studiano i “long term survivors” infetti da HIV ma senza i sintomi dell’AIDS e le terapie sono tuttora gli anti-retrovirali.
Come quelli di ieri sul CRISPR-Cas9, mi sembra un risultato utile perché è in parte negativo e non solo perché dimostra quanto siano diventati potenti i sistemi di screening.
